Automatic measurement is not affected by the surrounding environment. If the setting is always 0.5 mm or more, only the colonies are counted.

Solution

It is able to count colonies by size selection, so stable measurement without oversight is possible.

Reason

The diameter of a colony is calculated from its area value. And only the colonies with the set diameter are counted.
Since the threshold in binarization (sseparating colonies and others) can be fixed, only the colonies within the set diameter can be counted if the medium thickness is constant.

Even in the total number measurement, it may differ depending on the person how thin and small a colony is.
In automatic measurement, it can always be constant. In addition, the count can be corrected manually.
